This study evaluated the effect of prepubertal arsenic exposure in the liver and kidney of pubescent rats and their reversibility 30 days after arsenic withdrawal. Male pups of Wistar rats (21 days old) were divided into two groups (n = 20/group): control animals received filtered water, and exposed rats received 10 mg L-1 arsenic from postnatal day (PND) 21 to PND 51. The liver and kidney of 52 days old rats (n = 10/group) were examined to investigate the effects of arsenic on micromineral content, antioxidant enzyme activity, histology, and biochemistry parameters. The other animals were kept alive under free arsenic conditions until 82 days old and further analyzed by the same parameters. Our results revealed that 52-day-old rats increased arsenic content in their liver and arsenic and manganese in their kidney. In those animals, glycogen and zinc content and catalase activity were reduced in the liver, and the selenium content decreased in the kidney. Thirty days later, arsenic reduced the manganese and iron content and SOD and CAT activity in the liver of 82-day-old rats previously exposed to arsenic, while glycogen and selenium content decreased in their kidney. In contrast, PND 82 rats exhibited higher retention of copper in the liver, an increase in iron and copper content, and CAT and GST activity in the kidney. Significant histological alterations of liver and kidney tissues were not observed in rats of both ages. We conclude that arsenic-induced toxicity could alter differently the oxidative status and balance of trace elements in pubertal and adult rats, demonstrating that the metalloid can cause effects in adulthood.

Serious concerns have recently been raised regarding the association of Fe excess with neurodegenerative diseases in mammals and nutritional and oxidative disorders in plants. Therefore, the current study aimed to understand the physiological changes induced by Fe excess in Pistia stratiotes, a species often employed in phytoremediation studies. P. stratiotes were subjected to five concentrations of Fe: 0.038 (control), 1.0, 3.0, 5.0 and 7.0 mM. Visual symptoms of Fe-toxicity such as bronzing of leaf edges in 5.0 and 7.0 mM-grown plants were observed after 5 days. Nevertheless, no major changes were observed in photosynthesis-related parameters at this time-point. In contrast, plants growing for 10 days in high Fe concentrations showed decreased chlorophyll concentrations and lower net CO2 assimilation rate. Notwithstanding, P. stratiotes accumulated high amounts of Fe, especially in roots (maximum of 10,000 µg g-1 DW) and displayed a robust induction of the enzymatic antioxidant system. In conclusion, we demonstrated that P. stratiotes can be applied to clean up Fe-contaminated water, as the species displays high Fe bioaccumulation, mostly in root apoplasts, and can maintain physiological processes under Fe excess. Our results further revealed that by monitoring visual symptoms, P. stratiotes could be applied for bioindication purposes.

Knowledge of the pattern of exploitation of trees can help us understand the relationship between marmosets and plants, especially in declining forests, such as those in the Brazilian Cerrado. Black-tufted marmosets (Callithrix penicillata) regularly exploit exudates by gouging the bark of trees with their specialized teeth. Determining preferred tree species to exploit exudates is important for forest management aimed at maintaining an essential food source for urban marmosets. We characterized the tree species, dendrometry, and the characteristics of the holes made by marmosets to obtain exudates. Based on these data, we proposed a gouging effort index (Chiseling Suitability Index for Marmosets, ChiSI). We identified 16 species belonging to 10 families of trees with gouging marks made by marmosets. Eleven new tree species used by black-tufted marmosets for exudates were identified in urban forests in the Cerrado. Exudate exploration was predominantly of medium intensity, with round holes concentrated in the canopy. The species Tapirira guianensis and Croton urucarana were preferred. The ChiSI was characterized by a narrow range for both T. guianensis and C. urucarana. Despite the flexibility of obtaining exudates, the black-tufted marmosets concentrate their exploitation to only a few tree species. The T. guianensis and C. urucarana tree species should be considered the most important species for management plans and the preservation of black-tufted marmosets that live in urban forests in the Cerrado.

Plants used in phytoremediation should accumulate and tolerate a specific pollutant. Here, we aimed at evaluating a possible arsenic (As) accumulation and mechanisms of tolerance against As-induced damage in Landoltia punctata to explore this species for phytoremediation. Plants were subjected to increasing As levels. As absorption was higher with increasing As levels. The activity of superoxide dismutase and glutathione reductase as well as anthocyanin levels increased with As levels. Catalase and peroxidase activities increased in plants subjected to As levels up to 1.0 mg L-1 and decreased at higher levels. Due to the antioxidant system, higher levels of reactive oxygen species were restrained in plants under low levels of As. However, the levels of superoxide anion, hydrogen peroxide, and lipid peroxidation increased in response to the impaired antioxidant system induced by the highest As levels. Biomass decreased in plants exposed to As and scanning electron microscopy revealed root structural damage in the root cap of plants under 3.0 mg L-1 As. This work highlights that L. punctata can be considered a hyperaccumulator species and has potential for As phytoremediation when levels are lower than 1.0 mg L-1-a concentration 100-fold higher than that recommended for drinking water. Novelty Statement: Landoltia punctata can be considered a hyperaccumulator species and has the potential for arsenic phytoremediation when levels are lower than 1.0 mg L-1.

The objective of this study was to evaluate the concentration of potentially toxic elements in Brachiaria decumbens, Stylosanthes guianensis, and Saccharum officinarum plants and soil samples in affected and unaffected areas by rupture of the Fundão dam, Brazil. Samples were collected in areas affected by residues from the Fundão dam (RAA1, RAA2, RAA3) and in an unaffected area (control). The material was analyzed for the composition of micronutrients and trace elements in soil and plants, as well as contamination factor (CF), accumulation factor, and translocation factor (TF). Overall, the results showed that soil and plant tissues had increased Fe, Mn, Cu, and Cr content and decreased Zn content in the affected areas, compared to the control. Leaves and roots of B. decumbens showed an increase in Fe content in affected areas, compared to the control, reaching a mean maximum value of 42 958 µg/g of roots of RAA2-collected plants. As a result, CF for Fe of B. decumbens was classified as very high and they presented low TF values. Furthermore, B. decumbens collected in affected areas showed an increase of Fe, Mn, Cu, and Cr in leaves, stems, and roots, whereas in Stylosanthes guianensis, there was an increase of Fe concentration in all tissues and Cr in leaves. Also, Saccharum officinarum showed the accumulation of Mn in the stem and Cu in leaves and stem. On the other hand, there was no contamination of plants by hazardous elements such as Pb, Cd, and As in the samples analyzed. In conclusion, increases in the content of Fe, Mn, Cu, and Cr were found in soil and several plant tissues of residue-affected areas, which could compromise plant growth and represent potential hazards arising from the biomagnification process in the food chain. O objetivo deste estudo foi avaliar a concentração de elementos potencialmente tóxicos em plantas de Brachiaria decumbens, Stylosanthes guianensis e Saccharum officinarum e amostras de solos em áreas afetadas e não afetadas pelo rompimento da barragem de Fundão. As amostras foram coletadas em áreas afetadas por resíduos da barragem de Fundão (RAA1, RAA2, RAA3) e em uma área não afetada (controle). O material foi analisado quanto à composição de micronutrientes e elementos-traço no solo e plantas, além de fatores de contaminação (CF), bioacumulação e translocação (TF). No geral, os resultados mostraram que o solo e as plantas apresentaram maiores teores de Fe, Mn, Cu e Cr e menores teores de Zn nas áreas afetadas em comparação ao controle. Folhas e raízes de B. decumbens apresentaram aumento no teor de Fe nas áreas afetadas em relação ao controle, atingindo o valor máximo médio de 42.958 µg/g nas raízes de plantas coletadas em RAA2. Como resultado, CF para Fe de B. decumbens foi classificado como muito alto, mas com baixos valores de TF. Além disso, B. decumbens coletadas nas áreas afetadas apresentaram aumento de Fe, Mn, Cu e Cr nas folhas, caules e raízes, enquanto que em Stylosanthes guianensis houve aumento da concentração de Fe em todos as partes das plantas e Cr nas folhas. Saccharum officinarum também apresentou acúmulo de Mn no caule e Cu nas folhas e caule. Por outro lado, não há contaminação das plantas por elementos perigosos como Pb, Cd e As nas amostras analisadas. Concluindo, foram encontrados aumentos nos teores de Fe, Mn, Cu e Cr no solo e em vários tecidos vegetais, o que pode comprometer o crescimento das plantas e representar riscos potenciais decorrentes do processo de biomagnificação na cadeia alimentar. The microbial diversity and functioning around oceanic islands is poorly described, despite its importance for ecosystem homeostasis. Here, we aimed to verify the occurrence of microbe-driven phenanthrene co-oxidation in the seawater surrounding the Trindade Island (Brazil). We also used Next-Generation Sequencing to evaluate the effects of aliphatic and polycyclic aromatic hydrocarbons (PAHs) on these microbial community assemblies. Microcosms containing seawater from the island enriched with either labelled (9-14C) or non-labelled phenanthrene together with hexadecane, weathered oil, fluoranthene or pyrene, and combinations of these compounds were incubated. Biodegradation of phenanthrene-9-14C was negatively affected in the presence of weathered oil and PAHs but increased in the presence of hexadecane. PAH contamination caused shifts in the seawater microbial community-from a highly diverse one dominated by Alphaproteobacteria to less diverse communities dominated by Gammaproteobacteria. Furthermore, the combination of PAHs exerted a compounded negative influence on the microbial community, reducing its diversity and thus functional capacity of the ecosystem. These results advance our understanding of bacterial community dynamics in response to contrasting qualities of hydrocarbon contamination. This understanding is fundamental in the application and monitoring of bioremediation strategies if accidents involving oil spillages occur near Trindade Island and similar ecosystems.

Arsenic (As), in the form of trivalent arsenite or pentavalent arsenate, is a ubiquitous toxic compound naturally occurring in the environment. This study aimed to evaluate the impact of two different forms of inorganic As on reproductive parameters following oral exposure. Adult Wistar male rats were exposed to sodium arsenite or arsenate at concentrations of 0.01 mg/L or 10 mg/L for 56 d in drinking water. Sodium arsenite at both concentrations and sodium arsenate at 10 mg/L produced reduction in daily sperm production, in number of spermatids in the testis, and in sperm in the epididymal caput/corpus regions. Changes in epididymal morphometry were variable and region specific. Total and progressive sperm motility and sperm morphology did not differ markedly between controls and animals exposed to As. The body and reproductive organs weights, as well as testosterone concentration, remained unchanged among all groups. In conclusion, As exposure in drinking water over 56 d produced damage in male reproductive functions in adult rats, suggesting that fertility problems might occur. Therefore, additional studies need to be undertaken to investigate potential mechanisms underlying sodium arsenite- and arsenate-induced disturbances in fertility and reproductive performance.

We wished to evaluate the effects of arsenic on the morphology and anatomy of Brassica oleracea, Raphanus sativus, Brassica juncea, Brassica oleracea var. capitata and Brassica oleracea var. italica. Seeds were subjected to concentrations 0µM, 250µM, 350µM and 450µM arsenic in the form of sodium arsenate (Na2HAsO4·7H2O) during 12 days. All species accumulated more arsenic in the roots than in the shoots, except for B. oleracea var. capitata. There was no difference of translocation factor between species and treatments. Growth decrease was observed in roots of B. oleracea and R. sativus, and in shoots of R. sativus and B. oleracea var. italica. All species presented anatomical alterations in the roots, such as: cell hypertrophy, protoplast retraction, cellular plasmolysis, and necrotic regions. B. juncea presented collapse and hypertrophy of cells from the leaf blade tissues. Quantitative anatomical analyses performed on the root and leaves of B. oleracea and B. juncea revealed that arsenic interfered on the root vascular cylinder diameter and on height of epidermal cells of the adaxial leaf surface of both species. We concluded that arsenic was absorbed from the culture medium and induced alterations both on root and shoot growth of the seedlings. Retention of arsenic within the root was responsible for major damage in this organ.

Increasing contamination of aquatic ecosystems by metals has caused various morphological, physiological and biochemical changes in aquatic organisms, and the gills of fish are recognized as indicators of environmental quality. In this context, the present work proposed to study the effects of different concentrations of zinc (Zn) in the histology of gills of yellow tail lambari (Astyanax aff. bimaculatus) after acute exposure. Seventy-two adult males of A. aff. bimaculatus were used, the treatments were six concentrations of Zn: 0; 3; 5; 10; 15; and 20 mg/L of water, by 96 h, and gills, muscle and bone fragments were removed. Fragments of gills were fixed and included, sectioned in a rotary microtome and stained with toluidin blue. Fragments of bone, muscle and gills were dehydrated and digested to quantify the absorption of Zn. The median lethal concentration (LC(50)) 96 h after Zn acute exposure was 10 mg/L of water. Noteworthy, Zn was highly toxic in acute exposure trials starting at the concentration 5 mg/L. The exposure of fish to the metal caused branchial histopathological changes correlated with increasing concentration, caused the death of fish at concentrations of 10, 15 and 20 mg/L. The histological alterations observed in the gills were hyperplasia, lamellar fusion, aneurysm, destruction of the lamellar epithelium, rupture of membrane, deletion of secondary lamellar high, which presented more severity in treatments exposed to the highest concentrations. In conclusion, gills of A. aff. bimaculatus presented profound histological alterations as a result of Zn exposure, and hence, proved to be excellent indicators of environmental contamination.

RESUMO

This work aimed to study the partition of cheese whey proteins alpha-lactoalbumin and beta-lactoglobulin using aqueous two-phase system by applying the cloud point extraction technique. The cloud point temperatures were determined under different concentrations of copolymer and salt. The system providing the best protein separation conditions was 20 mass% of copolymer PE61 and potassium phosphate salt solution of 100 mM, at pH 7. The protein alpha-lactoalbumin remained preferentially in the aqueous phase and the beta-lactoglobulin was transferred to the copolymer phase.
